Partial purification of HA by organic solvents

Abstract
Hyaluronic acid (HA) has known as an important element in drug, biomedicine and cosmetics industries with a high commercial value due to its viscoelasticity, moisture absorption and high biocompatibility. The application and commercial value of this product is in relation with its purity. In this study, we used an appropriate solvent in order to remove existing impurities. The produced HA by a recombinant Corynebacterium glutamicume was precipitated with two solvents, ethanol and isopropanol with ratios of 1:1, 2:1 and 3:1 at 4 and -20 °C. The HA concentration, protein and nucleic acid were measured after the end of precipitation process. The results indicated that isopropanol in ratio of 1:1 at -20 °C had the highest amount of HA recovery, as 2.1 mg/ml. The mentioned solvent was more successful in removing impurities than others so that the amount of protein and nucleic acid were reduced to 0.052 μg/ml and 0.059 ng/ml, respectively.
