Optimization of the alkaline protease production by Bacillus licheniformis ATCC14580

The main objective of this study is the optimization of the alkaline protease production by Bacillus licheniformis ATCC14580 using the Taguchi statistical method. To do the effective factors on the production of alkaline protease including: temperature, glucose, yeast extract, glycine and pH were optimized at 4 levels by an L16 Taguchi orthogonal array. According to the result, glucose with 5 gl-1, yeast extract 15 gl-1, glycine 0.75 gl-1 and pH 9, soybean 40gl-1, KH2PO4/K2HPO4(7.6/24.8 gl-1), MgSO4.7H2O 0.2gl-1 and temperature of 50 °C were the optimized levels to obtain the highest volumetric productivity of protease. After optimization in the shake flask scale, 2638 IU.ml-1 and 164.5 IU.ml-1.h-1 were obtained for alkaline protease activity and productivity, respectively. The ANOVA showed the yeast extract and temperature were the most effective variables on the alkaline protease productivity.
